You can obtain a Nef rune through various in-game activities, including quest rewards, monster and treasure drops, and by transmuting other runes using the Horadric Cube.
Hellforge Quest Reward
One reliable way to get a Nef rune is by completing the Hellforge quest in Act IV. On Normal difficulty, there is a 1 in 11 chance for a Nef rune to be among the quest rewards that drop from the Hellforge.
Monster and Treasure Drops
Nef runes can drop from various monsters and treasure chests as you progress through the game. You'll start to find them with a higher probability from the Act 2 Good treasure class onwards. This means you have a chance to discover them from enemies and containers in Act 2 and all subsequent higher-level areas.
Horadric Cube Transmutation
The Horadric Cube offers a consistent method for crafting a Nef rune if you have the necessary components. By combining three lower-tier runes, you can transmute them into a single Nef rune.
